Lab 4: Culturing Escherichia coli involving Cav1.2, Connexin 43, and N-Cadherin By: Zachary Siou EGRB 491 Due: 2/28/17 Introduction Voltage gated channels are necessary components of life processes, in many organisms. One in particular, is the calcium voltage gated ion channel. Often lodged within the phospholipid bilayer, the imbalance of the calcium, or, the inside vs outside concentration, creates a gradient. The channel proteins often undergo conformations, states that which allow or block calcium ions from passing through. As ions move inside the cell, this creates a depolarization, or surge in the voltage. They form a network that provides an environment to foster proper growth and development. Again, this is widely expressed in cases of tumor growth in the area. Figure 1 [2]: The role of tubules in cell proliferation and di fferentiation of sperm cells. N-cadherin is a protein encoded by the CDH2 gene. It interacts with the cellular cytoskeleton, and is often involved in cardiac muscle, as well as certain cancers. Being calcium dependent, it helps to maintain cellular structure and integrity. For example, it plays a role in trans-endothelial migration, which involves cell-cell adhesion [3]. The endothelial layer contains many different fibers, as well as pathways that allow attachment for the cadherin protein. Some cancer cells can eventually pass through the endothelium, causing the cancer to become malignant and spread. Cadherins in this case can be used to identify and track the spread of the cells, and further identify common routes of travel through the human vasculature. The poem: â€Å"My Last Duchess† by Robert Browning gives a haunting look into the thoughts of a possible murderer. From the first reading one can deduce that the â€Å"Last Duchess† was murdered by the Duke. While the motive for the murder is not blatantly expressed, a deeper reading reveals that the Duke became aware of his wife’s rampant infidelity. This betrayal drove the Duke into a fit of jealous rage in which he murdered his wife in cold blood. So, â€Å"...twas not/ Her husband’s presence only, called that spot/ Of joy into the Duchess’ cheek...† (lines 13-15) is not an innocent expression describing a coy Duchess, brother a suggestive use of language that is employed by the Duke throughout the poem to allude to his his former wifes lack of†¦show more content†¦He states that he had difficulty discussing his worries about the Duchess’ fidelity with his wife. Perhaps this is a consequence of his noble upbringing. He admits however that if not for his wifes loose morals he would otherwise be sexually satisfied in his relationship. Yet as the Duchess’ flirtatious nature grew the ever more extreme. The Duke felt jealous, threatened, and that his wife was out of his Victorian era rule. He had enough, he had her ability to â€Å"smile† at other men put to an end â€Å"I gave commands; Then all smiles stopped together.†(lines 45-46). Presumably through having her die in some way, or by having the Duchess be put away. We do not know which due to such details being omitted by Browning for the sake of mystery. The Duke shows the wedding negotiator (the reader) a new piece of art, one of Neptune taming a sea-horse †Notice Neptune, though,/Taming a sea-horse, thought a rarity...†(lines 54-55). Theory: Many molecules absorb ultraviolet or visible light. The absorbance of a solution increases as attenuation of the beam increases. Absorbance is directly proportional to the path length, b, and the concentration, c, of the absorbing species. Beer’s Law states that A = ebc, where e is a constant of proportionality, called the absorptivity. Different molecules absorb radiation of different wavelengths. An absorption spectrum will show a number of absorption bands corresponding to structural groups within the molecule. In this experiment wavelength range between 400-650nm is used in order to detect in the visible range. Steel is an alloy primarily composed of iron with carbon content between 0. 2% and 1. 7% by weight. 1. 5% manganese is may be used too, to provide additional strength for a modest price increase. Carbon and other elements act as a hardening agent, preventing dislocations in the iron atom crystal lattice from sliding past one another. Manganese is essential to iron and steel production by virtue of its sulfur-fixing, deoxidizing, and alloying properties. In order to detect the manganese amount in the compound KIO4 is used to oxidize Mn+2 to Mn +7 since Mn+2 does not absorb in the visible region therefor cannot be detected. Tungsten lamp is used in the experiment to reach the range between 400-650nm. After the beam comes from the tungsten lamp, it passes through the monocromator to reach the wavelength range which was selected to find the maximum wavelength. After the light passes through the monocromator it goes through the cuvette and then the detector. The cuvette is made of quartz in order to pass the visible light whereas a glass cuvette would absorb it.
